美利堅合眾國第一夫人(First Lady of the United States of America,縮寫:FLOTUS),是美國白宮的女主人,通常是指現任美國總統的妻子。不過,若該總統是單身漢或鰥夫,或這總統的妻子未能勝任第一夫人的光環,第一夫人的職位就可能會改由總統的女性親戚或朋友擔任。

美國第一夫人不需要經過選舉,此身份亦無任何正式職責,亦無任何的薪酬或者津貼。僅是與總統,或者代替總統出席正式儀式。

傳統上,美國第一夫人不會保有其他工作,而且擁有下屬協助工作,包括白宮社交秘書(White House Social Secretary)、幕僚長、新聞秘書、首席花藝設計師(White House Chief Floral Designer)以及白宮行政總廚(White House Executive Chef)。

美國第一夫人辦公室也負責白宮的所有的社交和禮儀活動,是美國總統行政辦公室的一部分。
